A bio prac for substrate concentration should have been a part of your enzyme experiment. We did rennin and milk. For the substrate concentration, we had 1 test tube with 10ml of normal milk, a second test tube with 10ml of milk diluted with 10ml of water and in a third test tube we added 1 teaspoon of powdered milk. All of the test tubes were kept in a water bath at 37 degrees then we added rennin solution to all the test tubes and recorded the time taken for the milk to coagulate. The concentrated milk solidified quickly, the normal milk solidified normally, and the diluted milk didn't solidify. Hope that helps.

Another substrate experiment which we did was with Catalase (enzyme) and Hydrogen peroxide (substrate). collect 300ml of hydrogen peroxide. halve it, setting aside 150 ml and naming one beaker ' half concentration ' with the remaining hydrogen peroxide beaker add 150ml of water , name this beaker ' half concentration ' repeat step 3 & 4 until you have 5 different concentrations.
To test measure how fast the catalase paper rises to the top. good luck
