I have not actually done an exp on this but i guess you could observe plants for structures such as shiny or hairy leaves which reduce transpiration as the leaves would reflect sunlight and reduce air surrounding the leaf.
You could also look at things like if the leaves hang vertically such as in the mulga tree (leaves hang on a 45degree angle to direct rain/water towards the roots)
Also things such as small needle like leaves (thin which reduces exposure to sun hence reducing transpiration due to less surface area) An example is the Hakea tree
Theres also other biological things you could maybe look for with a microscope such as sunken stomates etc....
